Use Gigabyte sata or ICH8 sata

I am only setting up one 250gb hard drive on my new DS4 motherboard. Do I use the gigabyte sata or the ICH8 sata socket. How is it setup?. Also do I need to plug my 2x2 12v connector into the mainboard as well as the 24 pin atx connector.
